There isn't a file for his skin in the .jar of the game, nor anywhere else in the files. It is highly dubious and most likely a fan-made myth.

lol, i was just about to make a thread about this!!! i think he is real, but just a glitch. Also, Notch has no real brothers. He has one stepbrother who is still alive.

It was just a creepypasta and an ongoing joke for Mojang. There are no files relating to Herobrine in the vanilla game, and there never have been.

' Not entirely true.  Before Beta 1.8 there were files for the "Human" mob, which was the Steve skin. On lots of survival maps the makers would put these guys in there just as a little shock. That's about as close as it gets, though.

Yes, there was the "Monster" mob (aka the human), but you would need to change the texture of it to have white eyes for it to look like Herobrine. But yeah, I can see that happening.
